Quantity related problems
Successful sequencing is dependent on using the correct quantity of DNA template and primers.
Too little template
DNA template concentrations that are lower than those recommended result in low, noisy signal with no well defined peaks which is usually impossible to analyze.
Too much template
DNA template concentrations that are higher than those recommended result in pull-up peaks, which are multiple peaks in the same place. Pull-up peaks are caused when the very strong signal from the high template concentrations saturates the detector.
